The preventive effect of Bupleuri radix on Liver damage induced by acetaminophen in the rats

Abstract
Objective : Acetaminophen is a widely used analgesic and antipyretic drug throughout the world. It is also an over-the-counter drug and considered a safe drug. But its overdose and other causes such as alcoholism and diabetes can bring about hepatotoxicity. The hepatotoxicity of APAP is believed to be due to NAPQI produced by CYPs and reactive oxygen species (ROS). Method : Bupleuri radix(BR) at 500 mg/kg (p.o.) was administered to the male Sprague Dawley (5 weeks old)rats for one week, and then APAP at 500 mg/kg (abdominal injection) was given 24h after BR administration. This study was performed to research the protective effects of BR extract against APAP-induced hepatotoxicity. Result : The data including GOT, GPT, histological examination and GST indicated that BR has a significant antidotal effect on APAP overdose. Also, the date including LPO and catalase showed that BR has a antioxidant effect which is one of mechanisms of the hepatoprotective effect of BR. Finally, the data including GSH, GST, RT-PCR, Western blot showed that BR has a suppressive effect on the production of NAPQI, by regulating the expression of CYP 2E1. Conclusion : Based on these data, we concluded that the mechanisms underlying the protective effect of BR against APAP-induced liver damage are both the antioxidant effect against ROS and the inhibitory effect on the expression of CYP 2E1, which leads to the suppression of NAPQI production.
